What are zinc finger nucleases or ZFNs? How are ZFNs used for genetic/genome engineering and gene editing? How were these programmable nucleases engineered by combining zinc finger DNA-binding proteins with the FokI restriction enzyme cleavage or nuclease domain? In this video, I discuss the mechanistic basis of ZFNs, beginning with the structure and DNA recognition properties of C2H2 zinc finger proteins and the dimerization-dependent cleavage mechanism of the FokI nuclease. The video explains how arrays of zinc fingers can be engineered to target extended DNA sequences for genome editing. Finally, we cover the major limitations of ZFNs.
